In-vitro-synthesis of G-proteins into a Lipidmembran

The characterisation of diaphragm proteins under preservation of its natural and biophysical function makes high demands against the biophysical measuring methods. While soluble proteins can determine usually easily and be manufactured in larger quantities, diaphragm proteins without their hydrophobe environment lose fast their native folding and thus their function.

The diaphragm protein is the “Türsteher” or “Gatekeeper” of the cell. It steers, which may penetrate proteins or materials into the cell or leave them. A further function is a “guard”: If a protein, for example an body-own anti-body, docks at the free ends of the diaphragm protein outside of the cell on, this releases a signal cascade. This is the trip of a broad spectrum of physiological processes in the cell. In particular the group of the G-protein-coupled receptors (GPCRs) rank among the main targets of the active substance research.

The most difficult task with the investigation of GPCRs is the isolation in its natural form, without changing their characteristics. In aqueous solution they would clump immediately. The recognition of the ligands impairs substantially.

EH Kathrin Sinner succeeded at the institute for Max-Planck for polymer research in Mainz to develop a strategy the structure and characterisation one of the most important representatives of the class of the GPCRs made possible. It showed this by the example of the smell receptor OR5 of the rat (Rattus norvegicus). Its isolation and investigation in functional Proteinkonformation had so far not been possible.
Sinner brought together using cDNA as raw material in vitro synthesized GPCR with an artificial Lipidmembran. The two-layered Lipidschicht was applied on a gold sensor. The synthetic Lipidmembran copied the characteristics of a natural Lipidmembran. An arranged Insertion of the smell receptor protein actually took place into the peptid fixed Lipidmembran. Thus difficult cleaning or isolation process can be avoided.
